Physiological Metals Can Induce Conformational Changes in Transthyretin Structure: Neuroprotection or Misfolding Induction?

نویسندگان

چکیده

Transthyretin (TTR) is a plasma homotetrameric protein that transports thyroxine and retinol. TTR itself, under pathological conditions, dissociates into partially unfolded monomers aggregate form fibrils. Metal ions such as Zn2+, Cu2+, Fe2+, Mn2+ Ca2+ play controversial role in the amyloidogenic pathway. also present cerebrospinal fluid (CSF), where it behaves one of major A?-binding-proteins. The interaction between A? stronger presence high concentrations Cu2+. Crystals TTR, soaked solutions physiological metals Cu2+ but not Mn2+, Fe3+, Al3+, Ni2+, revealed an unusual conformational change. Here, we investigate effects have on order to understand if can induce specific active conformation guides its A?-scavenging role. capability certain accelerate process discussed.

Inhibiting transthyretin conformational changes that lead to amyloid fibril formation.

Insoluble protein fibrils resulting from the self-assembly of a conformational intermediate are implicated as the causative agent in several severe human amyloid diseases, including Alzheimer's disease, familial amyloid polyneuropathy, and senile systemic amyloidosis. The latter two diseases are associated with transthyretin (TTR) amyloid fibrils, which appear to form in the acidic partial dena...

Phosphorylated rhodopsin and heparin induce similar conformational changes in arrestin.

Photoactivated rhodopsin is quenched upon its phosphorylation in the reaction catalyzed by rhodopsin kinase and the subsequent binding of a regulatory protein, arrestin. We have found that heparin and other polyanions compete with photoactivated, phosphorylated rhodopsin to bind arrestin (48-kDa protein, S-antigen). This is shown (a) by the suppression of stabilized metarhodopsin II; (b) by cha...

Can mustard gas induce late onset polyneuropathy?

Abstract Background: Mustard gas, lethal in high doses, affects multiple organs such as skin, eye and respiratory system. We studied the development of late onset mustardinduced polyneuropathy among chemically wounded Iranian veterans. Methods: In this descriptive study, 100 chemically wounded Iranian veterans with severe eye involvement were examined for any signs and symptoms of polyneuropat...

3'-Azido-3'-deoxythymidine drug resistance mutations in HIV-1 reverse transcriptase can induce long range conformational changes.

HIV reverse transcriptase (RT) is one of the main targets for the action of anti-AIDS drugs. Many of these drugs [e.g., 3'-azido-3'-deoxythymidine (AZT) and 2',3'-dideoxyinosine (ddI)] are analogues of the nucleoside substrates used by the HIV RT. One of the main problems in anti-HIV therapy is the selection of a mutant virus with reduced drug sensitivity. Drug resistance in HIV is generated fo...

Neural populations can induce reliable postsynaptic currents without observable spike rate changes or precise spike timing.

Fine temporal patterns of firing in much of the brain are highly irregular. In some circuits, the precise pattern of irregularity contains information beyond that contained in mean firing rates. However, the capacity of neural circuits to use this additional information for computational purposes is not well understood. Here we employ computational methods to show that an ensemble of neurons fi...
